In the game, players try to stop balloons (called "Bloons" in the game) from reaching the end of a set route by placing towers or road items along it that can pop balloons in a variety of ways, often using monkey powers. Some towers can stop balloons and give other towers more time to pop them by freezing and gluing them together. Money is earned by popping balloons, completing levels, and collecting bananas from existing banana farms, which can be used to build new towers, upgrade existing towers, or temporary items such as exploding pineapples and road spikes.

The game is a tower defense game in which players can choose various types of towers and traps to place around the track to defend against balloons, and earn $1 in-game for each layer of balloons popped. If the balloon reaches the end of the path, the player loses lives (or, later in the game, health); once these are depleted, the game is over. Balloons always move along the map-set path on the track until they reach the exit, pop, or are moved to an earlier part of the track by the tower's ability.
